La Roja currently sit at the summit, though only on goal difference. Both they and Chile have four points apiece, with Mexico on two and Ecuador yet to get off the mark.
Victory for Chile would be ideal as it would allow the tournament hosts to remain in Santiago for the opening quarter-final, which takes place on June 24.
However, coach Jorge Sampaoli could yet be without star forward Alexis Sánchez for clash with Bolivia.
The Arsenal player was sent for an MRI after feeling pain in his right leg and it remains to be seen if he will be fit to feature on Friday.
Chile have scored five goals in their two fixtures so far, though Sampaoli will be disappointed they were so poor at the back in the 3-3 draw with Mexico.
Bolivia, meanwhile, will be looking to defeat their next opponents for the first time since 2000, when they triumphed 1-0 in a World Cup qualifier.
Confidence is high in the camp following a 3-2 victory over Ecuador last time out, Ronald Raldes, Martin Smedberg-Dalence and Marcelo Moreno (penalty) all finding the net.
Defender Edward Zenteno admits that La Verde are determined to upset the odds and claim top spot.
“We want to finish first in the group, we know it will be tough,” he told reporters.
“The aim is to do good things on Friday and go through as the best in the group.
“We are happy, but we will not lower our guard. We are mentally prepared to have a good performance in every game – the confidence is there.”
